Stem cell organoids are one of the most innovative tools in contemporary biomedical research, linking the conventional two-dimensional culture system to the in vivo animal model. Organoids are self-forming three-dimensional structures that resemble the structure and function of human tissue, and they are derived from either pluripotent stem cells (PSCs) or adult stem cells (AdSCs). They have quickly become invaluable in studying developmental biology, modeling diseases, regenerative medicine, and drug discovery. Unlike traditional cell culture, organoids restore the cellular heterogeneity and functional diversity of tissues and thus, for the first time, provide insight into both integrity and pathophysiology. This paper discusses the principles of stem cell organoids, their utility in different organ systems, and how they contribute to precision medicine, with a specific look at their use for modelling gastrointestinal disease.
Stem Cell Organoid Development
Organoids start from the induction of pluripotent stem cells (iPSCs), embryonic stem cells, or are grown from tissue biopsies carrying adult stem cells (AdSCs). Tang et al. (2022) describe how these cells can be channeled by using differentiation procedures in a step-by-step manner to create tissue-specific progenitors and self-assembly growth patterns into organoids. PSC-derived organoids have the advantage of pluripotency and, as such, can be differentiated into almost any cell type. In contrast, AdSC-Organoids offer greater short-term fidelity to the tissue of origin. Both methods produce systems that recreate the cellular makeup and the microarchitecture of organs such as the brain, liver, kidney, and intestine.
Applications Across Organ Systems
Organoid systems have now been demonstrated in a broad diversity of organ types. Tang et al. (2022) highlight advancements in brain organoids that can be used as models for neurodevelopmental disorders and infections, such as the Zika virus, and retinal organoids that provide models for degenerative diseases, such as retinal retinitis pigmentosa. Organoids, such as those from the kidney (for nephrotoxicity drug testing) and the liver (as promising targets for regenerative medicine and hepatotoxicity research), are widely used. Organoids for the heart and lungs also show promise for modelling congenital disease, viral infections, and regenerative medicine.
