Off-label and FDA-Approved Indications Acyclovir FDA-approved medication for Acyclovir is used to treat chickenpox, recurring bouts of genitals herpes simplex, and shingles (herpes zoster) indications (Li et al., 2021). In a person with an impaired immune system, preventing herpes simplex infection is an off-label indication. Cidofovir FDA-approved indication for this medication is the treatment of cytomegalovirus retinitis in patients with AIDs. The off-label indication is the treatment of additional infections caused by DNA viruses such as polyomavirus and adenovirus (Huntjens et al., 2023). Valacyclovir FDA-approved indications of Valacyclovir are the treatment of herpes zoster, the treatment of genital herpes simplex, and the treatment of cold sores. Treatment of adolescents contracting genital herpes again and suppressive medication that helps lower the likelihood of immunocompetence is the off-label indication of the medication (Li et al., 2021). Action Mechanism Acyclovir Action Acyclovir action mechanism involves it functioning as a nucleoside analog as it suppresses the production of viral DNA as it competes with guanosine triphosphate for the binding side of the DNA polymerase that causes a virus (Huntjens et al., 2023).
