Useful Antibiotics, Production of New Metabolite, and the Jumping Genes

Term Paper Instructions:

Read textbook or other literature to write a mini-review paper (at least one page for each question) to address questions. When you cite literature in your paper, please put the cited reference into your paper.

Your papers must be typed and double-spaced.

1. Most medically useful antibiotics interfere with either peptidoglycan synthesis or ribosome function. Why would the cytoplasmic membrane be a poor target for antibacterial medications?

2. You are a microbiologist working for a pharmaceutical company and discover a new metabolite that can serve as a medication. You now must oversee its production. What are some factors you must consider if you need to grow 5,000-liter cultures of bacteria?

3. Just before winter break in early December, your roommate stocks the refrigerator with a gallon of milk, but both of you leave before opening it. When you return in January, the milk has soured. Your roommate is annoyed because the milk was pasteurized and thus should not have spoiled. (1) Explain why your roommate’s position is unreasonable? (2) Design an experiment to prove that microbes do not spontaneously generate in milk?

4. As we know, Dr. Barbara McClintock (1902-1992) was a famous scientist who made several very important discoveries in genetics, and she received a Nobel Prize for her discovery of transposons, popularly called “jumping genes”. If you were asked to nominate her for the Nobel Prize, please search the internet and literatures, then make a statement supporting your choice.

Question 1: Most medically useful antibiotics interfere with either peptidoglycan synthesis or ribosome function. Why would the cytoplasmic membrane be a poor target for antibacterial medications?
The antibiotics ought to depict selective toxicity, where they can selectively destroy the bacteria system while not harming the host. Most antibacterial medications do not have a selective bacterial cytoplasmic membrane-targeting mechanism. Thus, such medications that are targeted at bacterial cytoplasmic membrane can equally damage cells in human systems, including the nervous system and kidneys. However, some antimicrobial agents directly target a part of bacterial cytoplasmic membranes in both Gram-negative and Gram-positive bacteria. These cytoplasmic targeting antibacterial agents are mainly cyclic peptides which have a rigid binding site capable of binding a lipid component. The cyclic peptides have binding sites that target bacteria, rather than being toxic to host cells (Epand et al., 2016).
Question 2: You are a microbiologist working for a pharmaceutical company and discover a new metabolite that serves as medication. You must oversee its production. What are some of the factors you must consider if you have to grow 5,000-liter cultures of bacteria?
During the process of the introduction of new antibacterial drugs, it is important to test its effectiveness by subjecting it to sensitivity and susceptibility tests. The cultured bacteria are exposed to such a new drug, which is the new metabolite in this case. A critical action entails a step is to grow different bacteria through a culture that will be used to examine the effectiveness of the new metabolite. The culture media is necessary for microbiological tests. However, it is essential to grow and obtain pure, uncontaminated cultures through proper growth, cultivation, and selection of microorganisms. Thus, it is recommended to be aware of factors or variables that interfere with bacterial culture growth. First, the quality and type of media affect the final culture outcomes. In the absence of quality culture media, the probability of getting accurate, predictable, reproducible, and repeatable tests becomes reduced. A microbiological culture medium represents a substance that facilitates the growth, support, and survival of bacteria. Culture media, among other things, should contain nutrients, growth-promoting factors, energy sources, buffer salts, minerals, metals, and gelling agents. Gelling agents are needed for solid medium.
There are typically two forms of culture media, which include liquid cultures, often called broth, and solid and semi-solid cultures, ordinarily called agar. There are further subtypes of the two media, for instance, growth media, which allows the growth of heterotrophic microorganisms, transport culture media that preserve such organism, enrichment media that works by increasing the cultured organism numbers, and selective growth culture media. The selective media will allow the growth of specific bacteria and restrict the growth of others. The pharmaceutical microbiology laboratory, like the one to test the efficiency of metabolite, deploys a given culture media depending upon the application required.
Some factors that affect bacterial growth and culture media need to be controlled. The growth of various microorganisms like bacteria is greatly affected by chemical and physical environmental conditions. The primary physical factors that influence growth in media culture include temperature, PH, oxygen levels, radiation, pressure, and solutes and water activity, osmosis. Based on temperature, bacteria can be divided into three categories for optimal growth. They include psychrophiles, which love cold, mesophiles that grow in moderate temperatures, heat-loving thermophiles, and hyperthermophiles, which exist in very high temperatures (Benammar et al., 2020). Based on their oxygen requirements, bacteria can be classified as obligate anaerobes, facultative anaerobes, aerotolerant anaerobes, obligate aerobes, and microaerophilic (Aslam et al., 2019). Depending on PH optimal survival values, there are categories of bacteria, including neutrophiles, acidophiles, and alkaliphiles (Thakur et al., 2020). Bacteria differ significantly on the ability to handle water activity relative to solute concentration.
During the culture of bacterial and the subsequent sensitivity and susceptibility test on metabolite, it is essential to ensure that the factors that affect the growth of each bacteria type are pr...
